Objective To investigate the effects of C.Spicatus on the kidney of diabetic rats, as well as possible the mechanism involved in.Methods After being injected with streptozotocin(STZ) the DM rat model was established. Rats were randomly divided into 4 groups. Group 1 was normal group. Group 2 was diabetic control group. Group 3 was given the C.Spicatus decoction at the dosage of 2g · kg-1· d-1. Group 4 was given the C.Spicatus decoction at the dosage of 8g · kg-1 ·d-1. After 8 weeks of administration, blood glycate, UAER, GFR of rats were determined respectively. The level of MDA, SOD, TGF-β1, FN, Col-IV, in the cortex of rats were also observed. Techniques including biochemistry, radioimmunosassay, spectrophotography, immuhistochemistry, light microscope, scanning electron- microscope.Results 1. 24h UAER of C.Spicatus group were significantly lower than those of diabetic control group (23.05 ± 2.53 μg · 24h-1) , (17.65 ± 1.76 μg · 24h-1 P < 0.01) in large dosage of C.Spicatus group, (19.29 ± 1.80 μg · 24h-1 P < 0.05) in small dosage of C.Spicatus group. 2. GFR of C.Spicatus group were significantly lower than those of diabetic control group (7.65 ± 0.61 ml · min-1 · kg-1), (4.97 ± 0.25 ml · min-1 · kg-1 P<0.01) in large dosage of C.Spicatus group, (6.60 ± 0.32 ml · min-1 · kg-1 P < 0.01)in small dosage of C.Spicatus group. 3. The level of SOD in the kidney of C.Spicatus group were higher than those of diabetic group (9.74 ± 1.04 U · mg-1 · prot-1) , (16.47 ± 2.10U · mg-1 · prot-1 P < 0.01) in large dosage of C.Spicatus group , (13.90 ± 2.00U · mg-1 · prot-1 P < 0.01) in small dosage of C.Spicatus group; while the level of MDA were higher than those of diabetic group (2.55 ± 0.18nmol · mg-1 · prot-1) , (1.82 ± 0.13 nmol · mg-1 · prot-1 P < 0.01) in large dosage of C.Spicatus group, (2.11 ± 0.16 nmol · mg-1 · prot-1 P < 0.01) in small dosage of C.Spicatus group. 4. The expression of TGF-β1, FN ,Col-IV, in the cortex of C.Spicatus group (8g · kg-1) were lower than those of diabetic control group (P < 0.05). 5. There were no significant differences in serum glucose between C.Spicatus group and diabetic group.Conclusion The C.Spicatus decoction at the range of dosage from 2g·kg-1·d-1to 8g·kg-1·d-1 has protective effect on the kidney of early diabetic nephropathy. The protective mechanism may be associated with the its improvement of oxidative stress and anti-inflammatory activity. In addition, it might relate to inhibitory effect on the proliferation of murine mesangial cells.
