Objective: The study was intended to verify Zheng's Plant Protein (abbreviation "ZPP") had tumor antigen activity. Researching the biodistribution of 131I labeling anti-ZPP-antibody tumor bearing mice, observing the tumor suppressive effect of the rabbit anti-ZPP-antibody and serum in vivo and detecting the expression the antibody in gastric tissue of normal person and patients,which were expected to provide experimental base for further studying the value of ZPP and anti-ZPP-antibody in the field of tumor immunodiagnosis and immunotherapy. Methods: 1.Chloramine-T is a kind of milder oxidizer which can oxide I - to I+. The latter can combine with hydrogen of the tyrosine residue under PH 7.5. The rabbit anti-ZPP-antibody labeled by 131I was prepared to the rule in view of the above, and the antibody was injected by tail vein in kunming mice suffered from S-180 tumor cells. Radioimmunoimaging (RII) and radioactivity of different tissue (eyeball blood,heart ,liver,spleen,kidney,muscle and tumor) were measured by γ-counter at 12h,24h,36h,48h,72h and 96h after injected. Then we calculated every radioactivity of unit quality and requested out its mean separately. Finally the %ID.g-1 (radioactivity uptake) and the T/NT (tumor tissue/not tumor tissue) were calculated. Therefore, we could study its biological distribution and radioimmunoimaging in the mice's organs and find if it could combine with the tumor cell significantly.2. S-180 is a kind of polymorphous sarcoma. Kunming mice were used as models. S-180 tumor cell dilution was injected by hypoderm in right armpit (0.2ml,each) respectively. The study objects were divided into three groups randomly (twelve mice every groups)—two experimental groups and one contrast group. Anti-ZPP-serum was injected in one experimental group through abdominal cavity, anti-ZPP-antibody was injected in another experimental group through coccygeal vein, and normal saline was injected in contrast group through abdominal cavity. Through that day, per three days once inject, the amount to be used was a 0.1ml to per mice each time. Watching entity the growth situation of the tumor day by day. Fifteen days after inoculating, we put mice to death by dislocating cervical vertebra, drew tumor tissue and weigh and then wrote down the results. Therefore we could calculate the tumor suppressive rates of the two experimental groups, and obtain the tumor suppressive effect of the rabbit anti-ZPP-antibody and serum in vivo. 3. The peculiar antibody which bringing the developing reagent can determine the nature and make a reservation and quantitative to the corresponding antigen at tissue/cell normal position through antibody-antigen reaction and the presentation color reaction of histochemisitry. According to the above principle Strptavidin-Peroxidasde(S-P) immunohistochemical technique was applied to detect the expression rabbit anti-ZPP-antibody in late gastric cancer ,early gastric cancer,gastric mucosal atypical hyperplasia,chronic gastritis and normal gastric mucosa . In order to prove ZPP had characteristic of human tumor antigen indirectly.Results: 1.Between 32~96h after tail intravenous injection, the image of the concentration radioactivity in tumor was obtained by the γ-camera and it was clearest during 48~72h. The higher concentration radioactivity place was consistent with the state of tumor. The rabbit anti-ZPP-antibody labeled 131I had a different distribution in the mice's organs and tumor tissue. The radioactivity uptake (%ID.g-1) of tumor was higher than other parenchyma organization (heart,liver,spleen, kidney,muscle) in different time points after injected the antibody, the %ID.g-1 of tumor tissue was 1.51±0.25,2.18±0.12,2.50±0.35,2.79±0.34,2.67±0.51 and 2.10±0.46 respectively in 12 h,24 h,36 h,48 h,72 h and 96 h after injected the antibody. But the %ID.g-1 of other tissue only reached 0.98±0.18 except blood. Radioactivity concentration of blood was higher than other tissue. The radioactivity ratios of tumor tissue/not tu...
