Cervical cancer is one of the most frequently seen cancer of gynecological cancers,in recent years the incidence of cervical cancers is increasing and young women is more frequently involved than before. The trauma brought to the patients on the body and mind by this disease and the metastasis ^ bad prognosis are still waiting to be solved. All these necessitate it for us to know more about the mechanism , biological characteristic and new treatment. Recent research shows that angiogenesis, the formation of new blood vessels, is required for the growth and expansion of tumours and the study on the positive regulator and negative regulator of tumor angiogenesis is becoming another hotspot.Cervical cancer is also angiogenesis dependent. Endostatin is an endogenous angiogenesis inhibitor that was first isolated from the supernatant of an in vitro culture of EOMA cells ,a murine hemangioendothelinma cell line. Further characterization revealed that endostatin is a 20-kd C- terminal fragment of collagen X VIII.In in vitro culture human and murine endostatin specifically inhibit the proliferation and migration of capillary endothelial cells and can induce apoptosis of proliferating endothelial cells. VEGF(Vascular endothelial growth factor,VEGF) is a glycoprotein of about 45 kDa and consists of two identical peptide chains linked bydisulphide bridges, it is known to be the most important pro-angiogenic factor, critical to the process of angiogenesis. It can increase vascular permeability, which may facilitate tumor dissemination via the circulation, It may also inhibit endothelial cell apoptosis by inducing expression of the survival gene BCL-2. VEGF appears to play key roles in the maitenance of normal situation and the occurrence and development of tumor.now there is still no report on the expression of endostatin and VEGF in cervical cancer and its clinal meanings. Objective: This study aimed at investigating the expression of endostatin and VEGFin cervical cancer and its clinical meaning in order to elucidate their roles in tumorigenesis and progression of cervical cancer,so as to provide a potential new way for prognosis assessment and give us a new idea on the treatment of cervical cancer.Materials and methods:1 .52 cases of cervical cancer samples(including 43cases of squamous cancer,9 cases of adenocarcinoma) and 14 cases of normal cervical tissue samples were collected. The tissues were fixed in 10% formalin and embedded in paraffin. No patients had received preoperative chemotherapy ,radiation therapy or other biological therapy.2.1mmunohistochemical streptavidin peroxidase method was used to detect the expression of endostatuu VEGF in all samples.3.The results were analyzed by SPSS10.0 software wrap.using Chi-square, Fisher's exact test, analysis of variance(ANOVA) and t-test.Statistically significant level was considered as "alpha equals 0.05".Results:l.The positive staining of endostatin protein is mainly observed in the cytoplasm of cancer cell, endothelium cells of blood vessel, and its positive rate is 70. 04 2 1.23%. in normal cervical tissue the positive rate is only21.43 3.18%.the result show a significant difference between the two groups(P<0.05),the positive rate of endostatin in highly ,moderately and poorly differentiation of cervical cancers is 66.13+7.72%; 79.56+3.37%; 92.08+2.02% respectively, and a statistical significance is observedbetween highly and poorly differetiation of cervical cancers(P<0.05),the expression ofendostatin is increased in FIGO 0 , I and IK IIIand IV. a significant difference isfound between 0 I and IK III and IV (PO.05) ,and the positive rate of endostatinin the team with pelvic lymph node metastasis is much higher than in the team withno pelvic lymph metastasis (P<0.05) .2.The positive staining of VEGF protein is mainly observed in the cytoplasm of cancercell.the positive rate of VEGF protein in cervical cancer is 59.6%, there is nopositive staining in normal cervical tissue.the result show a significant differencebetween...
