Abstract/Summary:
Stem cells are multipotent cells, which can replicate as undifferentiated cells and that have the potencial to differentiated to lineages of tissues. these stem cells exist in fetal and adult tissuthe, the former are called embryonic stem cells , the latter are called adult tissue stem cells. In the past, we thought adult tissue stem cells only can been found in hematopiesis and epithelium. With the technology developing of isolation and culture, stem cells can be isolated from hepatic neural, pancreas, adipocytes, muscle, which prove that adult tissues stem cells generally exist.In 2000, Gronthos isolated a clonogenic, rapidly proliferation of cells from adult human normal impacted third molars' dental pulp, who raised the notion of dental pulp stem cells (DPSCs) permanent teeth. in this study, we isolated dental pulp cells from casted. deciduous teeth by way of Friedenstein's method .we showed that most of the cells were positive stained by FITC labeled CD44,CD105,CD166,CD29,FLK-1 ,and only 1% of cells were positive for CD31,CD45,HLA-DR,CD11a,CD34.we found culture expended deciduous teeth DPSCs could be directed into the odentoblast lineage as detect by DSPP. We also found that this DPSCs could give rise to the heama land the neural lineage.To Keep the live pulp and to promote its repairto maintain the construction of the tooth have been what many scholar work hard for many years. Odontoblast,vessel and neural are the most important part. This research directed DPSCs differentiate into odontoblast lineage, heama and neural lineages which made it sure that it is possible to regenerate the tooth using DPSCs.Generally think, Mensenchymal stem cells (MSC) cultured in vitro with small a large nucleus and scant cytoplasm which physical volume is small, and report the shuttle type.MSC didn't express antigens CD14, CD34, CD45, but expressed antigens SH2,SH3,CD29,CD44,CD71,CD105,CD166. And other surface proteins. This study, we isolated dental pulp cells from casted. Deciduous teeth by way of Friedenstein's method. These cells'morphology and phenotype were similar to Mensenchymal stem cell. So, we think they along to Mensenchymal stem cell. Since 1867 Cohnheim isolated MSC from bone marrow, men have made lots of achievement in isolating, culturing, Biology characteristic, divide potential, the gene treatment. MSC derived from different tissue have comparability in character and Plasticity in differentiation, the found of MSC from dental pulp is not only to provide the new esource for MSC, also to provid the DSPP the theories foundation with the technique support into the research.Plasticity is that issue specific stem cells could differentiate into lineages other than the tissue of origin. The microenvironment supports the fate of stem cells. If the DPSCs belong To MSC really, then according to Plasticity the stem cells, DPSCs equally having the plasticity, can under the certain term divide into the other cells from the other tissues. In addition, deciduous teeth can be take easily, Can avoide immunity rejection. We believe it will provide the seed cell for tissue engineering, the aspect will have the vast foreground.
