Objective:To studay the changes of apoptosis of renal cells at rats during acute ceretral ischemia and relation with Ca2+ , tumor necrosis factor(TNF) and malondialdehyde(MDA).To observe the action mechanisms of renal damage in ischemia cerebrovacular disease. Methods:36 Wistar rats were randomly divided into the control groups(n=6) and the ischemia group .The ischemia model of rats (n=30) was divided 5 groups of 30min, 60min, 90min, 120min and 180min by ligation of right common carotid artery and cogalation the right vetebral artery. The following was obtained:the level of Ca2+, TNF-α and MDA in plasma and renal tissue .At the same time the number of renal cells apoptosis were also determined. Results: (1)The level of BUN and the level of Cr at 180min showed a notable discrepancy compared to control group and 30min group (p<0.05). (2) The number of renal apoptosis was higher in experimental groups than in control group (p<0.01); The number of renal apoptosis at 90min , 120min and 180min showed a notable discrepancy compared to 30min and 60min group (p<0.01). (3) The content of MDA at 60min, 90min, 120min and 180min were significantly elevated compared to normal control between in plasma and in renal tissue (p<0.05-0.01 ). The content of MDA had a positive correlation between in plasma and in renal tissue (r=0.81. p<0.01). (4) The content of Ca2+ in plasma at 90min, 120min and 180min were higher than the control group (p<0.05). The content of Ca2+ in renal tissue at 60min, 90min, 120min and 180min were higher than the control group (p<0.05-0.01). (5) The level of TNF-α inplasma at 60min, 90min, 120min and 180min showed a notable discrepancy compared to control group (p<0.05-0.01). TNF-protein level at 180min were significantly elevated compared to normal control and at 30min in renal tissue ( p<0.05-0.01 ) . (6) Alterations of renal ultrastructure were seen under election miscroscope.Conclusions: The level of Ca2+ MDA and TNF-a in plasma and in renal tissue were all. elevated progressively,which induced renal cells apoptosis through signal conduction. And the decreasing of renal function was consistented with renal pathological changes.
