Objective To study the expression effect of recombinant HBV containing IFN-a gene and X-gene defect in HepG2 cell line.Methods pcDNA3-KN-FlF2-IFN-a , the HBV expression plasmid containingIFN-a gene and X-gene defect, and three kinds of control plasmid were transfected into HepG2 cells. Then, fiuorescence PCR, RT-PCR and ELISA were used to detect the products.Results A system, HepG2/KN-FlF2-IFN-a, which could constantlyproduce, recombinant HBV containing IFN- a gene and X-gene defect , was set up successfully; in contrast with control virus, the level of KN-F1F2 -IFN-a were lower in duplication. So did the expression of HBsAg; both IFN mRNA and IFN protein were detected.Conclusions The recombinant HBV containing IFN-a gene and X-genedefect could be packaged in this way ; IFN- a could constantly be produced in HepG2 cell mediated by the recombinant HBV; it was possible that HBV might be reformed as a gene transfer vector.
