Development And Identification Of Monoclonal Antiboby For Detecting Plasmodium Sporozoite | | Posted on:2005-03-23 | Degree:Master | Type:Thesis | | Country:China | Candidate:R J Zhang | Full Text:PDF | | GTID:2144360125468376 | Subject:Pathogen Biology | | Abstract/Summary: | | | The current work is to obtain monoclonal antibodies (McAbs) of Region II+ of circumsporozoite protein (CSP) and fructose 1,6-bisphosphate aldolase (ALD) of a key ATP-generating enzyme in the glycolytic pathway, and to evaluate the possibility of using the antibodies in detecting of plasmodium sporozoites in mosquitoes.BALB/c mice were immunized with 12 peptides within Region II+ in CSP of P. falciparum. 3 positive hybridoma cell lines were obtained. ELISA results indicated that the McAbs could react with recombinant CSP fragment containing conserved Region II+. IFA showed that the McAbs could recognize the sporozoite of not only P. falciparum, but also P. yoelii. But the reaction with P. yoelii was poor than the reaction with P. falciparum.The ALD encoing gene was amplified by PCR from genomic DNA of FCC1/HN strain. ALD is expressed in E. Coli as a soluble form. BALB/c mice were immunized with purified aldolase protein developed strong immune responses to the antigen. We obtained 7 positive hybridoma cell lines. ELISA results indicated that the McAbs could react with recombinant aldolase protein. IFA showed that 4 lines McAbs can recognize the cultured P. falciparum, but not other 3 lines McAbs.
