| Genomic DNA were respectively extracted from surgically separated normal tissue,cancer-surrounding tissue, cancer tissue including breast cancer(3 cases) > breast benign tumor(l case) ,lung cancer(l case) , gastric carcinoma(l case) by using traditional hydroxybenzene-chloroform method .Then we amplified these DNA with RAPD by using 8 random primers ( SI , S4K S80, S8K A-01 , A-02 , A-03, A-04 ) . PCR products from RAPD were electrophoretically separated in agarose gel to detect DNA polymorphism-Relationship between each primer ar d DNA polymorphism of RAPD PCR products in each case was analyzed.The results show: For most of the primers a multiband profile of RAPD PCR products was seen in 6 case of tumors. Most of the primers produced amplified products that were moaomorphic. The minority produced amplified products that were polymorphic and many differential bands were acquired-Morever, DNA polymorphism could be detected in all of the malignant tumors,while DNA polymorphism hadn't been detected in benign tumors.These results suggested the genomic alteration could be detected in malignant tumor tissue by randomly amplified polymorphic DNA analysis(RAPD). Associated differential bands could be used for seeking tumor- associated genes further.RAPD te:hnology also could be used in early diagnosis and prognosis evaluation of tumor clinically. |
