Dividing them into 3 groups by random: delivery collagen membrane FK506 group, FK506 dropping group (control group 1) and delivery collagen membrane CSA(control group 2). (1) Detecting the drugs concentration and lasting period by ELISA on 1, 2, 4, 8, 24, 72 hr after surgery. (2) Observing the extent of immunologic rejection in corneal and the concrescene and pathologic changes of epithelial. (3) To evulate the immunologic rejection rate of corneal grafting during different times and detect the ultrastructure of corneal by electrical microscope in the average 3 follow-up.Results: The concentration in corneal,conjunctiva and aqueous humor in delivery collagen membrane FK506 group is higher than that in control groups. The concentration in corneal in delivery collagen membrane FK506 group is the highest and about 5 times of controls. The concentration in aqueous humor in delivery collagen membrane FK506 group after 72 hr is still 10 times of the concentration of control groups after 8 hr. There are few corneal lympocyte experimental group ,whereas lympocytes and acidophilic cells are abundant in control group .Transmission analyse by electon microscop indicates that cells in experimental group are conjugated closely ,and nucleus of corneal ephithelial cells in which chromatin is distributed erenly are elliptic and flat.In the same time organellase are rich in plasma and microdowns on cell are visible also. However,ultramicrostucture of cell in control group has already dissolved. Conclusions: The delivery collagen membrane FK506 can obviously increase and sustain the concentration of FK506 in corneal, conjunctiva and aqueous humor, and effectively suppress the immunologic rejection after surgery.
