Objective To study the expression and the significance of STAT3,cyclinD1 and Bcl-xl in primary hepatocellular carcinomas (HCC). Metheds Expression of STAT3, cyclin D1 and Bcl-xl proteins weredetected by immunohistochemistry technique (SP) with 50 cases ofhepatocellular carcinomas who had undergone surgery for HCC, 20 casesof their pericarcinomatous tissue, and 10 cases of normal liver tissue ascontrol specimens. Results â‘ The average positive expression of STAT3 in HCC is 45.42±24.23%, which is significantly higher than that in the pericarcinomatoustissue (15.96±8.47%, P<0.01) and normal liver tissue (8.96±2.26%,P<0.01). There is significant difference between the expression of STAT3in the normal liver tissue and that in the pericarcinomatous tissue, as well 6é‡åº†åŒ»ç§‘大å¦ç¡•å£«å¦ä½è®ºæ–‡(P<0.01). The strong positive expression of STAT3 in HCC gradesâ… â…¡and grades ⅢⅣare 41.94% (13/31) and 63.16% (12/19), respectively, andthere is no significant difference between them (P>0.05). The strongpositive expressions of STAT3 in HCC TNM stagesâ… â…¡and stages ⅢⅣare 41.38% (12/29) and 61.90% (12/19), respectively, and there is nosignificant difference between them (P>0.05). â‘¡The average positiveexpression of cyclinD1 in HCC (35.59±23.45%) increases significantlythan that in the pericarcinomatous tissue (11.79±6.38%) and normal livertissue(8.32±1.27%)(P<0.01, respectively). The strong positive expressionsof cyclinD1 in HCCs gradesâ… â…¡ and grades ⅢⅣ are 22.58% (7/31) and52.63% (10/19), respectively, and there is significant difference betweenthem (P<0.05). The strong positive expressions of cyclinD1 in HCC TNMstagesâ… â…¡ and stages ⅢⅣ are 20.69% (6/29) and 52.38% (11/21),respectively, and there is significant difference between them (P<0.05). â‘¢The average expression of Bcl-xl in HCC (38.33±23.36%) increasessignificantly than that in the pericarcinomatous (11.34±5.20%) and normalliver tissues (8.06±1.74%)(P<0.01, respectively). The strong positiveexpressions of Bcl-xl in HCC grades â… â…¡ and grades ⅢⅣ are 38.71%(12/31) and 42.11% (8/19), respectively, and there is no significantdifference between them (P>0.05).The strong positive expressions ofBcl-xl in HCC TNM stagesâ… â…¡ and stages ⅢⅣ are 37.93% (11/29) and 7é‡åº†åŒ»ç§‘大å¦ç¡•å£«å¦ä½è®ºæ–‡42.86% (9/21), respectively, and there is no significant difference betweenthem (P>0.05). â‘£The expressive intensities of cyclinD1 and Bcl-xlproteins are positively related to that of STAT3 in HCC. The correlationcoefficients are 0.69 and 0.80 (P<0.01), respectively. The expressiveintensities of cyclinD1 and Bcl-xl in the pericarcinomatous tissue arepositively related to that of STAT3, as well. Their correlation coefficientsare 0.55 and 0.54 (P<0.05), respectively. The expressions of them have nocorrelationship in the normal liver tissue (P>0.05). Conclusions (1) The expression of STAT3 is upregulated obviously inHCC and pericarcinomatous tissue, indicating the effect of STAT3 on HCCprogression. (2) The expressions of cyclinD1 and Bcl-xl in HCC increaseabnormally, indicating the effect of them on HCC progression. Theexpression of cyclinD1 has relationship to malignancy and prognosis, aswell. (3) As the target genes of STAT3, the positive correlations betweenthe expression of cyclinD1 and Bcl-xl and that of STAT3, indicateprobably that the mechanism of HCC occurrence is some target oncogenesof STAT3 to be aberrant upregulated, as the result of constitutive activationof it. This would result in abnormal proliferation and anti-apoptosis ofhepatic tumor cells.
