"stress" or "stress reaction" is a kind of general physical and adaptive response during the body is stimulated by a stressor. Nervous system, endocrine system and immune system are the main systems to maintain the body's internal milieu stabilization. Stress reaction is nervous system's activities, and there is obvious changes in endocrine system and immune system, too. Conduction center-hypothalamus of nervous system plays a very important role in stress reaction, it has a short-time effect on stress by means of stimulating sympathetic nerve and adrenal medulla; has a long-time effect on stress by means of stimulating pituitary anterior lobe and adrenal cortex to change the level of the hormone. Stress action is profit to the body , in general, there is correlation between the degree of and stress and stimulation level of stressor. Selye said, "adequate stress is profit to the body", as follows, a long-time movement training can improve the ability of space learning memory and can enhance the activity of cleaning free radical system. But, when stressor is overstrong , the body can not response adaptively because of the body's complexity, which leads to stress disturbance. Once the body is in a state of stress disturbance , the body is secondary lesion and may result in death, therefore overstrong stress is a hot spot of researching stress; there is individual difference in secondary lesion.To investigate the exact relationship between the two items, I detected the genetic polymorphism of 53 KM mice with the technique of Polymerase Chain Reaction (PCR) and measured Charcoal Clearance Index against the antigen during innate immune response by Charcoal Clearance Assay(CCA) and the content of cortisol in the serum by radioimmuneassay(RIA) . Therefore the second exon of H2-Eb was chosen for the detection of allelic polymorphism and the charcoal clearance index. Based on the nucleotide sequences of the second exon of Mus musculus domesticus (western European house mouse), 2 pairs of PCR primers (namely MudoEb5, MudoEb7) were designed, each having one sequence-specific primer. The primer sequences of internal positive control were derived from murine tumor necrosis factor alpha (TNFα). There were 64 positive cases of PCR in total (27 cases were from MudoEb5 and 37 from MudoEb7) as all the internal controls were successfully amplified, that is to say, 60% of alleles (64/106)were determined. Moreover, 17 cases were both positive for MudoEb5 and MudoEb7 detection. Besides, The data show that there is obvious decline inαof the restraint stress group compared with the control group(P﹤0.005); but there is not obvious increase in cortisol of the restraint stress group compared with the control group(P﹥0.005) and there is significant decline inαof the heat stress group compared with the control group(P﹤0.05); there is significant increase in cortisol of the heat stress group compared with the control group(P﹤0.05);.. There is little relation between MudoEb5 and stress immune and there is close relation between MudoEb7 and stress immune. Different types of stress haves strong suppressive effects on the immune system, and degree is linked with genetic polymorphism in KM mice.
