Objective To investigate the clinic diagnostic significance of CD44v8-10/CD44v10 and the relations between it and clinical pathologic factors by detecting expression of CD44v8-10 in primary lung cancer on mRNA level.Methods Competitive reverse transcription-polymerase chain reaction (Competitive RT-PCR) was performed to amplify CD44v8-10 and CD44v10 in primary lung cancer at the same time, and Gel-Image Analysis System was used to assay expression quantity of CD44v8-10 and CD44v10 respectively. By calculating the ratio of CD44v8-10/CD44v10 in lung cancer group and comparing with controls, to build a accurate molecularobiological indicator which used in verdicting malignance or benign of lung lesion.Results 1) Strong expression of CD44v8-10 was detected in primary lung cancer tissues, whereas it was very weak or no detectable in lung benign lesion and matched normal lung tissues; 2) The ratio of CD44v8-10/CD44vlO in primary lung cancer tissues was over 1.00 (3.78 1.98) compared with under 1.00 (0.43 0.18) in benign control, so the dividing point of CD44v8-10/CD44v10 between malignant and benign lesion was 1.00; 3) Significant correlations were not observed between CD44v8-10/CD44v10 ratio and clinic pathological features such as ages, sex and tumor histopathological classification (t test, P>0.05); 4) The ratio of CD44v8-10/CD44v10 in patients with lymph node metastasis was strikingly higherthan in those with no lymph node metastasis (t test, P<0.001); the ratio of CD44v8-10/CD44vlO in patients at stage II and III was significantly higher than those at stage I (t test, P<0.001).Conclusions The ratio of CD44v8-10 was a very fine indicator to verdict malignance or benign of lung lesion. The value of CD44v8-10/CD44v10 was more bigger, the risk of lymph node metastasis was m ore higher; otherwise the value of CD44v8-10/CD44v10 was more smaller, the risk of lymph node metastasis was more lower. The values of CD44v8-10/CD44v10 may be a significant clinical indicator in the evaluation of the stage and patient's prognosis and the potential tendency of lymph node metastasis.
