Objective To explore the influence of IgG-antisperm antibody in seminalplasma on the sperm acrosome reaction (AR). Methods ELISA tests were performed in our study to select theIgG-antisperm antibody positive semens.The semens then were centrifuged andthe seminal plasma were isolated,30 such samples were collected.Spermatozoafrom ten healthy donors were incubated with those samples;they were comparedto a control group of 10 samples without antisperm antibodies and 10 sampleswith buffer simultaneously.The spontaneous acrosome reaction (SAR) and theinduced acrosome reaction (IAR) by progesterone were determined by means ofa flow cytometer.The FITC-PSA was used to stain the sperm acrosome ,whichcan radiate green fluorescence at 510~550nm wavelength as excited by 488nmwavelength laser.The value of FITC fluorescence was accumulated andcalculated by a special computer as the stained sample passed the flowcytometer,the results were output as numeric value. Results The spermatozoa treated with IgG-antisperm antibody positiveseminal plasma showed significantly lower IAR responses than the two controlgroups(p<0.001),but there was no significant difference between control groupsin IAR(p>0.05) ;and there was no significant difference between the threegroups in SAR(p>0.05). Conclusion IgG-antisperm antibody from immunologic infertilitypatients is able to inhibit progesterone induced acrosome reaction.
