Objective: Infection of Helicobacter pylori (H.pylori) existed prevailingly,which was closely related to upper digestive tract diseases. Oral cavity was the second colony of H.pylori besides stomach. The existence of H.pylori in oral cavity was harmful to both stomach and oral health. So it was significantly important to study effective drug to get rid of H.pylori in oral cavity. In this study, inhibition of gargle with Chinese herbal medicine on H.pylori was observed by polymerase chain reaction(PCR) and the clinical effect and application value of gargle with Chinese herbal medicine were evaluated to provide an effective way to get rid of H.pylori infection in oral cavity. Methods:The compound prescription was composed of rhizoma coptidis,phellidendri ,sweeten,flavoring and antiseptic. After soaked in water for two hours rhizoma coptidis and phellidendri and suitable water were heated twice and filtrated. Flavoring was also heated in the same method. All the filtrated fluids were mixed in proper proportion. Sweeten and antiseptic were added. The final mixture was stored for use after filtrated.160 testees were selected with the standard of acceptance as follows: (1) no severe general systematic diseases. (2) no administration of antibiotics and drug of treating stomach diseases within a month. (3) no periodontal therapy within six months. The saliva of these testees were detected by PCR. Sixty H.pylori positive patients were chosen and divided into two groups randomly,the test group and control group, each 30. Gargle with Chinese herbal medicine was used to rinse by the test group for 7 days, three times a day,10~15ml,3 minutes each time. After rinsing, eating and rinsing were prohibited within 30min. 0.9% saline was used to rinse by the control group in the same method. After 7 days saliva of the test group and control group was collected and detected by PCR: (1) the extraction of DNA (2) the identification of DNA (3) PCR amplification (4) The PCR products were analyzed by gel electrophoresis and the results were recorded. The data were analyzed by SPSS software.Result:1. Sixty H.pylori positive patients were selected from 160 testees ,and the H.pylori positive rate was 37.5%. 2.There was no significant difference in age and sex between the test group and control group (pï¹¥0.05). 3.Detected by PCR, the H.pylori negative rate of the test group was 53.85% after treatment, while the H.pylori negative rate of the control group was 4.00%. There was significant difference between the test group and control group (p<0.01). 4.No side effect was found with using gargle with Chinese herbal medicine presently.Conclusion:1.PCR was an ideal method to detect H.pylori in oral cavity. 2.Gargle with Chinese herbal medicine, which was proved clinically killing H.pylori and having no local stimulation, was developed. 3.Gargle with Chinese herbal medicine containing mainly rhizoma coptidis and phellidendri was effective to kill H.pylori in oral cavity.
