Objective The research was designed to observe the therapeutic effect of Dakang Capsules on erectile dysfunction (ED) animal models and investigate the possible mechanism. We hoped it could offer experiment proof for the therapy principia of promoting blood circulation to relieve stasis on ED patients. Methods We used the normal and suspensory irritability mic., emasculated infancy rats and aged rats as objects respectively to observe how the drug influenced the mating function of the normal and suspensory irritability mice. the growth of sexual organs of the emasculated infancy rats. the quantity of NO in serum and the penile cavernous tissue . related sex hormone(T.FSH, LH). the erectile function and the quantity of Nitric oxide synthase(NOS) and its sub-type(iNOS and cNOS) in the penile cavernous tissue of aged rats. Results The drug could increase the mating function of the normal and suspensory irritability mice(the latter P<0.05)and accelerate the growth of sexual organs of the emasculated infancy rats(the group of high dosage and the index of the foreskin gland were marked increased, P<0.05);It decreased the testosterone level in serum of the emasculated infancy rats (P<0.05 ) ;Compared with the hormone level of emasculated infancy model rats, FSH and LH of the groups administered by the drug increased, and the group of mid-dosage increased obviously(P<0.05); The drug enhanced the quantity of NO both in serum and the penile cavernous tissue; It could shorten the erectile latent period of aged rats (P<0.05) and increase the erectile intensity (P>.05); Compared with the controls, the quantity of t-NOS. iNOS in the penile cavernous tissue of aged rats increased significantly (P<0.05); Conclusions The experiments were certified the universality and validity of the drug in treating animal ED models. As a non-libido intensifier, it didn't affect the level of male hormone in serum, which accords with the status quo that the majority of ED patients don'tlack male hormone. More investigation showed that, the effect might be due to the enhancement of t-NOS and iNOS in the rat's penis cavernous tissue, especially the direct influence of iNOS on NO-cGMP signal passageway, which may be the key tache.
