As a kind of part-anesthetic drug, lidocaine was chemically manufactured in 1920s,but its extensive and specific pharmacological effect, especially the dual effects on central nerve system has been carefully investigated in recent ten years. This article has discussed its effect on several epilepsy models and studied its acting mechanism. The results are as following.1 Successfully made the amygdala kindling model of rats.2 Lidocaine 5.0-25 mg kg-1 ip can inhibit the seizure of kindled rats, raise the threshold, reduce Racine's stages (P<0.05). Lidocaine, at the dose above 30 mg kg-1, shows the significant excitory effect on kindling.3 When combined with valproate at an ineffective dose 50 mg kg-1 ip, lidocaine can reduce Racine's stage (P<0.05).4 Successfully made the lidocaine chemical-kindling model, discussed the relation of chemical-kinding and doses of lidocaine, drug-administration plans, animal species.5 Studied several kinds of anti-epilepsy drugs on lidocaine-induced chemical-kindling model.6 In electricity-stroke model of mice, lidocaine 20-40 mg-kg"1 ip can significantly prolong latent period(P<0.05).These results show, that lidocaine has excitory and inhibitory effects on amygdala kindling models and has synergic effect with valproate, the mechanism probably is related to enhancing the function of GABA system and inhibiting Na+channel. Lidocaine is a kind of chemical-kindling agent, its mechanism and characteristic are different form general drugs(such as PTZ). These results provide the basis of lidocaine's pharmacological mechanism and reasonably drug-administration, especially combined administration of anti-epilepsy drugs; alsodo some help to the study of epileptic pathogenesis .
