Objective To compare hemolytic activity of crude venom of Stomolopus nomurai which obtained by method A with crude venom which obtained by method B. To study on the acute toxinology of crude venom of Stomolopus nomurai which was administated to mice via i.v.Methods (1)Aliquots of 1ml of a 0.5% human red blood cell suspension were incubated overnight at 4 C with different protein equivalents of crude nematocyst venom(81.6ug/ml , 102.4 u g/ml, 128.0 u g/ml , 160 u g/ml, 200 u g/ml, 250 u g/ml )which obtain by method A., the dose of crude venom which obtained by method B were 98.3 u g/ml , 122.8 u g/ml , 153.8 ug/ml , 192 u g/ml, 240 ug/ml, 300 ug/ml respenctively. Aliquots of lml of a 0.5% human red blood cell suspension were incubated with equivalens of 10mMPBS(PH 7.4) and 10mMPBS(PH 7.4) added to 0.1mg/ml saponin. The concentrations of the later present in the supernatant were determined spectrophotometrically at 545nm. The lytic potency of the tested venoms was expressed as % hemoglobin released compared to the total hemoglobin present after maximal lysis.(2) Divise 100 mice into five group of 2.700mg/kg, 2.160 mg/kg, 1.728 mg/kg , 1.384mg/kg , 1.106 mg/kg by random. Each mouse was injected into 0.5ml crude venom via tail vein.and observed amount of the dead mice with 7days, and record every organ reaction, then autospy the dead mice. The mice brain, liver and lungs were fixed for histological exmination in 4%. The specimens were embedded in paraffin stained with hemotoxylin and eosin, and observed under light microscopy. Results (1) The hemolytic activities were no different between crude venom which obtain by methods A and crude venom which obtained by method B. (2)The crudevenoms are hemolytic to human erythrocytes and ED50 values: 151.89 ug/ml.(3)The crude venoms are lethal to mice and LD50 values: 1.6mg/kg.There was abnoaml in mice brains, livers and lungs under light microscopy.Conclusions These results indicate that the crude venom was hemolytic to human erythocytes and lethal to mice .
