Objective: To investigate the formate toxicity on retinal Muller cell and the neurotoxicity quantity-efficacy relation. Methods: Retinal explants obtained from 10 neonatal rats were primary and passage cultures, and gained Miiller cell .The forth Generation were implanted into a piece of 96-well culture plates.The 96 wells were divided into two groupds: control group and formate (5mM l0mMN .20mM. 30Mm 50mM. 100mM and 200mM) exposure group. The effects of formate on culture rats retinal Miiller cell were examined by MTT and LDH release and morphology observation. Results: Histopathologic changes were apparent in retinal Miiller cell after formate intoxication, with evidence of inner segment swelling and mitochondrial disruption. Formate exposure 24hr group: at the 5mM l0mM concentrations of formate, the OD and LDH release in retinl Miiller cell had no difference from that the control group (p>0.05). At the concentration of formate more than and equal 20 mM , the OD and LDH release in retinal Miiller cell was significantly less than that of the control group (p<0.05), and with the increase of formate concentration. Conclusion: 1.Formate are able to demonstrate its histopathologic changes neonatal rat retinal Muller cell when acquiring certain concentration after 24hr incubation in vitro, retinal edema and vacuolation in Miiller cell , swelling and disruption of the mitochondrin. 2. Cytochrome oxidase is the terminal component of the mitochondrial electron transport chain and is an integral protein complex of the inner mitochondrial membrane. This enzyme participates in the reduction of oxygen to water with the consequent synthesis of ATP. Formate inhibits the activity of cytochrome oxidase , then inhibits retinal energy metabolism. 3.Formate has the neurotoxicity for retinal Miiller cell in vitro, and the effect is dose-dependant.
