ObjectiveThere are three stages in the transplantation of the pancreas including warm and cold ischemia and ischemia/reperfusion process. The susceptibility of the pancreas to ischemia/reperfusion injury has been demonstrated in experimental studies and in clinical settings such as transplantation of the pancreas. Ischemia/reperfusion injury appears to be one of the important factor in various of complication after transplantation of the pancreas. The participation of the proinflam-matory cytokine have played an important role in ischemia/reperfusion injury. Various cellular protective agent added to the solution to reduce ischemia/reperfusion injury is one of the aim in transplantation. Taurine (2 - aminoethanesul-fonic acid) the most abundant free amino acid in the body, is a free beta - ami-no acid that is normally present in high concentrations in many tissues of humans and animal species, Derived from methionine and cysteine metabolism, taurine is known to play an important role in numerous physiological functions, including modulation of calcium levels, maintenance of osmolarity, anti - oxidation, and stabilization of membranes. Taurine also has beneficial effects in ischemia/ reperfusion - injury by preventing the production and release of proinflammatory cytokine. In this experiment, we have observed the change of serum amylase and histopatholgy and we have slso observed the change of TNF - a and IL - 1 in pancreas after transplantation through the model of rat pancreas - duodenal transplantion. We have studied the influence of taurine on TNF - a and IL - to reveal the anti - edema and Cellular protective mechanism of taurine. so the preservation and function post - transplantation can be improved.Materials AND MethodThirty Wistar rats (200 -250g purchased from the experimental animal department of China Medical University) were studied. Rats were randomly allocated into three groups (n =10) , including control group (CMU -1 solution) experiment group A ( CMU -1 + TAU 5mM ) , experiment group B ( CMU - 1 + TAUlOmM). The pancreas was transplanted to the recipient after preservation for six hour. Every group was divided into two subgroups according to reperfu-sion time. Namely blood and pancreas were collected at the time of 2h and 6h after reperfusion to inspect blood amylase and TNF - a and IL - 1 (3. Taking the pancreas organizations to make the pathologic check. Passing the light microscopic examination, in control group hyperemia and edemathe can be checked in mesenchymal of pancreas two hours after transplantation and companied with hemorrhage of tisse, the gland acinus suffers to press and atrophy and the gland alveolus cell dies. Inflammation becomes to aggravate gradually along with time. In same time limit the pancreatids is obvious alleviative, Inflammation does not becomes to aggravate along with reperfusion in the experiment group A than that in the control group. There is not much apparent difference between experiment group A and experiment group B. TNF - a and IL - 13 are expressed in pancre-as tissue two hours after reperfusion. The expression of TNF -α and IL - 1 β were increased apparently along with time. The taurine can reduce the expressions of TNF - α and IL - 1β at the same time limit. ( P < 0. 05).
