This condition usually lead to heat stroke, heat hyperpyrexia or heat collapse, and symptoms such as headache ,eclampsia, examination. Scholars have pay close attention to the procedure, mechanism, therapy of such injury and hope to solve the problem of the prevention, therapy and rehabilitation. So the study of the heat stress response not only have hygiene significance but also have prophylaxis significance.From bacteria to human, heat stress can lead to heat stress response. The behavior of this response is cause a series of polypeptide-heat stress proteins, which is a protein play a protective role for human. It is known to all that human can develop adaptation after heat stress, and heat adaptation can reduce the psychological tension caused by heat stress, and raise the thermotolerance ability, but heat adaptation have a limt A excessive adaptation usually lead to heat injury, so study the procedure, mechanism, biochemical event of heat stress response have active significance in scientific research and clinical practice. On the one hand ,the results of heat stress response can cause the increased synthesis of heat stress proteins; necrosis and apoptosis of cells and a series alteration on morphology andat molecul level. On the other hand, the increased synthesis of heat stress proteins have protective function for the outcome of the heat stress. In the stage of heat stress damage, a series of sub-cellular and molecular pathological changes in cells of damaged regions have also taken place, which could cause the damage of cell membrane, organs and DNA. The damages of DNA might be the result of many factors as well as be the reason of many new changes in cell. More and more studies have showed that apoptosis caused by DNA damage participated in the pathological course of the heat stress response and became one of the main causes of the heat stress response. Apoptosis is an active, manipulative and normal phenomenon, which is regulated by a series of proteins. This research can provide a new method to prevent apoptosis of neurons caused by heat stress response and try to find a new protective agent involving in apoptosis. At present, many studies have showed that hyperthermia can lead to many cell apoptosis, but only a little report based on primary cultured neurons. The purpose of our study is to investigate the morphological changes of apoptosis and cell vital force of neurons in order to understand the biology alteration after hyperthermia injury, hope to find out some protective agent during hyperthermia.In this experiment, we established neuron hyperthermia injury model via primary cultured neurons. Divide the identified mature neurons as normal group and hyperthermia injury group. At each time point, we observed morphological changes by invert microscope, HE stain and Klenow Method to detect DNA breaks. Results in the experiment were shown as following:[1] Optical microscope indicates: We can observe that normal nerve cell is round shape after digestion by trypsin; 10 hours later, cell body become Round or ellipse, and the dispersed cell stick to wall partly; 24 hours later cell body become fusiform or triangle, most cell adhere to wall, and apophysis have elongation; most cell have two apophysis, and can seegrowing tip; 3 days later, apophysis is growing continuely and bond together as neural network; 7 day later, cell of control group have light areola, and neural network is dense; hyperthermia group have increase in drift cell; the number of neural cell decrease; neural network became sparse ; apophysis of some cell were drifted; some cell lost their apophysis.[2] HE stain indicates: We can observe mat the cytoplasm of neuron is in pink color and the nucleus is light blue under optical microscope.
