| Objective To study the treatment effect of Qingning, a kind of Chinese herbs compound, on rats with experimental liver fibrosis and the influence on the expression of TGF- 1 and PDGF-B mRNA, to explore the possible mechanism of Qingning's anti-liver-fibrotic action .Methods 168 SD rats were randomly divided into two groups: the normal and the experimental. Rats with experimental liver fibrosis were induced by injecting 40%CCl4 3ml/kg body weight subscutaneously twice a week for nine weeks, on the first time the dosage was double. Rats in the normal group were given the same dosage of saline by injecting subscutaneously. After the model being made, rats in the experimental group were divided into the liver-fibrotic model, the big, the middle, the small volume of Qingning and the colchicine groups. Rats in the big, the middle and the small volume of Qingning groups were given 22g/kg-d, llg/kg-d and 5.5g/kg-d of Qingning, rats in the colchicine group were given 0.5mg/kg-d of colchicine, rats in the normal and the liver-fibrotic model group were given the same volume of saline, once a day for nine weeks. All the rats were killed at the end of the eighteenth week. The severity of inflammation and fibrosis in all liver tissuses were measured bysemiquantitative scoring. The expression of TGF- P i and PDGF-B mRNA in all liver tissues was detected by in situ hybridization method.Results (1)The severity of inflammation and liver fibrosis scorings in rats of the liver-fibrotic modle group were obviously higher than those of the normal group (P<0.01); after the rats had been treated by Qingning and colchicine, the severity of inflammation and liver fibrosis scorings in rats of all the treatment groups became obviously lower than those of the liver-fibrotic modle group, which showed significant and very significant(P<0.05, P<0.01); the severity of inflammation scoring in rats of the big volume of Qingning group was higher than those of the middle, small volume of Qingning group and the colchicine group, which showed significant and very significant (P<0.05, P<0.01), but the liver fibrosis scoring of all the treatment groups showed no significance(P>0.05). (2)The expression of TGF- 1 and PDGF-B mRNA in liver tissues of the liver-fibrotic modle group was significantly higher than that of the normal group (P<0.01); The expression of TGF- 1 and PDGF-BmRNA in rat liver tissues of all the treatment groups was obviously lower than that of the liver-fibrotic modle group, which showed very significant. (3)There was positive correlation between the expression of TGF- 1 mRNA and the liverfibrosis scoring in the liver-fibrotic modle group; the same relationship exists between the expression of PDGF-B mRNA and the liver fibrosis scoring in the liver-fibrotic modle group.4Conclusions (1) Qingning can relieve of the severity of inflammation and liver fibrosis in rat liver tissues. (2)TGF- β1 and PDGF-B mRNA in liver tissue of liver-fibrotic rats can express increasedly, TGF- β1 and PDGF-B relate with the happening and developing of liver fibrosis closely. (3) Qingning can decrease the expression of TGF- β1 and PDGF-B mRNA, which might be a mechanism of the Qingning anti-liver-fobrotic action. |
