Antifibrotic Mechanism Of Compound Formula 991 Kangxian Tang

Objective: To study the antifibrotic effect of compound formula 991 Kangxian Tang on experimental hepatic fibrosis in rats and explore the mechanism.Methods:The experimental hepatofibrosis rats were produced by CCL4,and meanwhile were treated with compound formula 991 Kangxian Tang(991KXT) or colchicine(Col) as control group. At the end of experiment, the rat liver pathological changes were observed, the amount and distribution of vimentin for the detection of hepatic stellate cell(HSC) were immunohistochemically assessed. Also malon- dialdehyde(MDA) and superoxide dismutase(SOD) in the liver, alanine aminotransferase(ALT), albumin (ALB) and hyaluronic acid(HA) levels in the serum were measured .Results:1. In the 991KXT group, the serum level of ALT were decreased markedly and the serum level of ALB were raised significantly, Showing a significant difference in comparison with those in the model group(P<0.01). 2. The level of serum HA in the 991KXT group was significantly lower than that of the model group (P<0.01) and the col group (P<0.05). 3. The MDA content was markedly decreased in the 991KXT group as compared to the model group and the Col group. 4. 991KXT improved the liver injurious and fibrotic pathology. Immunohistochemistry indicated that 991KXT reduced the percent area of vimentin positive cells in liver and the effect didn't exist in Colchicine.Conclusion: Compound formula 991 Kangxian Tang has significant antifibrotic effect for experimental hepatofibrosis in rats, one of the mechanism may beassociated with it can inhibit the proliferation of HSC by antiperoxide injury of hepatocyte.
