Objective To study the effect of adenosine preconditioning against ischmia-reperfusion injury to lung.Methods The in vivo rabbit ischemia model with occlusion of left pulmonary hilus for 45 minutes was established in this experiment. Seventy-two white rabbits were divided into three groups. Group I (n=24):no ischemia reperfusion ;Group II (n=24) : ischemia-reperfusion;Group III (n=24):adenosine intravenous injection at 10 minutes before ischemia-reperfusion .The changes of left lung wet-dry weight, malondialdehyde, myeloperoxidase and expression of the CD11b/CD18 on PMN by flow cytometric analysis in the injury lung were measured in each group at the time of 1 hour, 2 hours , 4 hours reperfusion after 45 minutes of pulmonary ischemia.Results In group II ,partial pressure of oxygen in artery decreased evidently in 2 hours and 4 hours after reperfusion . Left Lung wet-dry weight ,the contents of malondialdehyde , myeloperoxidase were increased evidently after reperfusion (P<0.05) ; the expression of CD11b/CD18 were upregulated. In group III ,the changes aboved were improved as compared with group II.Conculsion Adenosine preconditioning prevented ischemia-reperfusion lung endothelium injury and improved postischemia-reperfusion lung function by inhabiting neutrophil sequestration and activation in lung.
