The antiinflammatory effects of the glucosides of cheanomeles speciosa (GCS) were studied by acute and chronic inflammatory models(the models of carrageen induced paw swelling in rats and cotton induced granuloma in rats )and type II collagen-induced arthritis( CIA).The effects and mechanisims of GCS on inflammatory and immune reponses were studied in vivo and vitro.The intragastric administration (ig) with GCS (120 , 240mg 'kg-1) could significantly inhibit carrageen induced claws swelling in rats and GCS (60 , 120 , 240mg kg-1) inhibited cotton embedding method induced granuloma in rats. These results showed that GCS had anti-inflammatory effect on acute and chronic inflammatory models.The onset of paw swelling was on d24 after injection of chicken collagen type II, the swelling peak appeared on d36-40 and then declined after d40.It was found that GCS had prophylactic and therapeutic action on CIA in mice . GCS(60, 120, 240 mg kg-1,ig) could significantly inhibit secondary inflammatory swelling and polyarthritis, reduce the elevated PGE level located in ankle and pain scores in inflamed paws. These results suggested that GCS had antiinflammatory and analgesic effects.Thymus index and spleens index were assayed on d44 after immunization. Results showed that spleens index of CIA mice was increased, thymus index was not changed , GCS (120 , 240mg kg-1) reduced the elevatory spleens index of CIA mice. T,B cell proliferation in CIA mice were increased, GCS (60 , 120, 240 mg 'kg-1)could suppress the elevated proliferation of T cell and B cell to ConA andLPS respectively . The production of IL-1 by PM4>and IL-2 by T cell were upregulated significantly in CIA model. GCS(60, 120, 240 mg 'kg-1) decreased the production of IL-1 and IL-2.The level of serum anti- CII antibody was increased significantly in CIA model. GCS(60, 120, 240 mg kg-1)could reduced the level of plasma anti- CII antibody.Pathological change and X-ray change in ankle of CIA mice showed that the synovium of CIA was hyperplastic, synovial cells were increased ,pannus was formed and inflammatory cells infiltrated into synovium. Hyperplastic synivium .inflammatory cells infiltration, and pannus formation were significantly reduced in CIA after administration of GCS.All theses results suggested that GCS had antiinflammatory ,analgesic and has immunomodulatory effects.
