Objective:There are a few medicaments can be sellected in the chemotherapy plan on oral carcinoma.Taxol is a crude drug of anti cancer,it has remarkable effects on many different cancers.Taxol has an unique function.There are fewer basic studys on oral carcinoma in our country.By the studying of the noxious effects of taxol on tongue carcinoma cell Tca8113 and Adenoid cystic carcinoma cell Acc2 and the studying of the effects of induce apoptosis,to offer the reference for the clinic chemotherapy.GSH(Glutathione) is the most important no-protein hydrosulfide group,it has manny important physiological function. L-Buthionine-S-R-Sulfoximine is the inhibitor of the enzyme to compose glutathione.The objective is to study the enhanced sensitivity of L-Buthionine-S-R-Sulfoximine to taxol.Materials and Methods:By the technic of cell culture,within the enviroment of laboratory ,under different consistence of taxol,to study the noxious effects of taxol on tongue carcinoma cell Tca8113 and Adenoid cystic carcinoma cell Acc2 using the technic of MTT,and to study the effects of apoptosis by TdT-mediated dUTP nick end labeling and Flow CytoMeter,and to value the effects of taxol on oral carcinoma.To evaluate the influence of L-Buthionine-S-R-Sulfoximine on taxol within the same thing of not changing the other condition in the same laboratory.Results: Different consistence of taxol can evidently restrain the grows of tongue carcinoma cell Tca8113 and Adenoid cystic carcinoma cell Acc2.The group of high consistence of taxol can restrain the grows of cell distinctly in a short time and has an upper ratio of apoptosis.The highest ratio of apoptosis is 34.1 5.8%.The groups of midst and lower consistence of taxol also have good effects.To prolong the exposure time ,the groups of midst and lower consistence of taxol had an obvious value. L-Buthionine-S-R-Sulfoximine has a feeble effects on Tca8113 and Acc2.It has slender noxious effects under the consistence of 0.01, 0.1,0.5,1mmol/L,24h pretreatment of L-Buthionine-S-R-Sulfoximine has no effects on taxol.Conclusions:Taxol has distinct effects to block the grows of tongue carcinoma cell Tca8113 and Adenoid cystic carcinoma cell Acc2,and has high ratio of apoptosis.Its effects has positive correlation with consistence and time.The factor of timer has a higher effect.To prolong the exposure time,the effect would be prominent.The effect of taxol on two cells has no different.L-Buthionine-S-R-Sulfoximine has feeble noxious effects,pretreatment of L-Buthionine-S-R-Sulfoximine has no effects on taxol.The effects of taxol are not dependent on glutathione, L-Buthionine-S-R-Sulfoximine can not enhance the effects of taxol.
