ObjectiveTo evaluate the analgesic effect of Tianyuan Ketong Prescription on differnent pain model, and explore its mechanism; To provide theoretic support for clinical applying and development of new form of Tianyuan Ketong Prescription, and enrich the theory of traditional Chinese medicine on pain management.Methods1. The acetic acid-induced body-writhing mouse pain model, radiant heat stimulation- induced tail flick rat pain model, and formalin rat pain model were used to observe analgesic effect of Tianyuan Ketong prescription;2. Immunohistochemistry technique was used to study Tianyuan Ketong prescription's influence on c-fos expression and content of SP in superficial lamella of spinal cord dorsal horn of formalin pain model rats; radioimmunoassay was used to study Tianyuan Ketong prescription's influence on contents of PGE2 in local inflammtory tissue and blood plasma and that of -EP in blood plasma of formalin pain model rats.Results1. Tianyuan Ketong prescription could notably decrease times of body-writhing induced by acetic acid in mouse (P<0. 05), and prolong latency of tail-flick induced by radiant heat stimulation in rats(P<0. 05);2. Tianyuan Ketong prescription could obviously inhibit painreactions in formalin pain model rats, decrease the release of PGE2 ( P < 0. 05 ) , and suppress effect of enhancement of c-fos expression and of increase of SP' s content;3. Tianyuan Ketong prescription could obviously improved the state of increase of Blood viscosity caused by inflammtory reaction in formalin pain model rats (P<0. 05);5.Tianyuan Ketong prescription could notably suppress the state of decrease of P -EP in blood plasma cused by pain reaction in formalin pain model rats.Conclusions1. Tianyuan Ketong prescription possesses obvious analgesic effect on acetic acid-induced body-writhing mouse pain model, radiant heat stimulation- induced tail flick rat pain model, and formalin rat pain model;2. Decreasing release of PGE2, suppressing expresson of c-fos and generation of SP, improving blood circulation, and increasing content of β-EP in blood plasma may be the Key links of analgesic effect of Tianyuan Ketong prescription.
