Through observe the changes in structure and neurotransmitter of operation sample from epilepsy Cerebral cortex spike wave focus, to find out the nosogene-sis of epilepsy with view of Nerve - chemistry and morpholog Conjugation.MethodsPatient selection: Tissue samples of epilepsy Cerebral cortex spike wave focus ( n = 28 ) with typical epilepsia disorder discharge from VEEG and brain tissues ( n = 12) were obtained from patients of Cerebral ventricle tumors in the neurosurgery department of 463 Hospital of liberation army between March, 2003 and, Octobe 2003.Histology: Sample fixation, routine desiccation and paraffin embedding, at last HE staining.Immunohistochemistry ; Paraffin sections were cut at a 7 -μm thickness, after repairing antigen and blocking nonspecific reactivity, the sections were incubated with anti - Glu monoclonal antibody for one night at 4℃ thermostat and then with biotin - conjugated second antibody for an additional 20 min. Positive signals were detected by the S - P method with S - P kit.electron microscope Observe: Sample fixation, Cleaning with bi - distilled water,gradient desiccation with Alcohol and Propylene oxide,resin embedding to get together, Staining with Heavy metal, Observe and Photograph with ransmis-sion electron microscope.Result determination:The labeling indexs of Glu were calculated microscop-, ically field at a magnification of 400 x by counting male cells from 5 selected fields, to indicate with x s.Statistical methods: student t - test was used.ResultsWe find aniso - degree disease alteration in epilepsy Cerebral cortex spike wave focus through histology and electron microscope, nerve cell concintration, Nucleus deformation and nuclear membrane Shrinkage, mitochondrion swelling and Ribosomal Depolymerization, Lysosome and lipofuscin increasing. Presyn-aptic edema with Round synaptic vesicle Aggregation, synaptic space Widening. Cytoplasm of Astrocyte edema, Nucleus Shrinkage, endothelial cell of blood capillary swelling, myelin sheath Brisement, breakage, to demyelination. nerve cell has Glu - Immunologic reaction, glial cell proliferation, the most is micro-glia, and Proliferative glial cell has Glu - Immunologic reaction too. There are significant difference between result and contrast with t -test, p <0. 01.Conclusions1. we find aniso - degree Disease Alteration in epilepsy Cerebral cortex spike wave through histology and electron microscope about nerve cell , Presyn-aptic , Astrocyte, blood capillary and myelin sheath, stellate cell proliferation.2. nerve cell has Glu- Immunologic reaction, glial cell proliferation, the most is microglia, and Proliferative glial cell has Glu - Immunologic reaction too.3. Epileptic attack is not only temporary brain functional disorder caused by simple alteration of physiological and biochemical, it is important for the changes of spike wave focus form constitution and neurotransmitter to cause epilepsy.
