Objective: The aim of the study was to detect the DNA of Ascaris lumbricoides in the intrahepatic stone using molecular biological method ,thus to provide a potent approach for the etiology study of intrahepatic stone and clues for the primary prevention of this disease. Methods: 1 DNA was Extracted from Intrahepatic stones as template . 2 Identifying the target region of Ascaris lumbricoides and the primers :ITS-2 sequence was selected because it is a species-specific marker of this group of parasites. 3 DNA from Intrahepatic stones was amplified with the specific primers and subjected to electrophoresis and sequencing analysis. Results: There were 12 positive results in 56 cases of Intrahepatic stones, that were confirmed by subsequent sequencing analysis. Conclusions: There was DNA of Ascaris lumbricoides retaining in a certain proportion of intrahepatic stones, which indicates that Ascaris lumbricoides may play a role in the formation of Intrahepatic stones.and the detailed mechanism need further investigation.
