BackgroundOvarian carcinoma is one of gynecological malignancies with poor prognosis, epithelial malignancies of the ovary remain confined to the peritoneal cavity even in advanced stages of the disease, it has been suggested that the growth of ovarian carcinoma intraperitoneally could be related to the local deficiency of antitumor immune effect mechanism, allowing tumor growth within the peritoneal cavity. But,up to now. the mechanisms responsible for peritoneal immunodeficiency of ovarian epithelial carcinoma(OEC) remain unclear. One possible mechanism is the abnormal expression of cytokines in tumor region. Recently years, study shows cytokines maybe play important roles in the development and growth of cancer, including ovarian carcinoma.Interleukin-10(IL-10)is a potent anti-inflammatory cytokine that preferentially acts on the cell-mediated(THl-like) immune response,which is generally regarded as the adequate immune reaction against malignant tumors. IL-10 thus interferes with tumoricidal mechanisms,it suppresses secretion ofTNF- a and of reaction oxygen intermediates in macrophages,downregulates MHC I expression on tumor cells and interferes with accessory cell function of antigen-presenting cells. On the other hand, IL-10 inhibits the production of IL-2 and INF- v by TH1 cells,it also inhibits the proliferation of human peripheral blood T cells, and the synthesis of INF- Y and other cytokines in human peripheral blood mononuclear cells(PBMC). Moreover, IL-10 has also been shown to inhibit antigen presentation by macrophages and Langerhans cells, which makes tumor cell escaping immune assault . Abundant expression of IL-10-specific mRAN and IL-10 protein has been detected in malignant ascites, serum and within tumor tissue. The question of which cell population is responsible for the production of IL-10 in ovarian carcinoma has so far been addressed solely by the investigation of ovarian carcinoma cell lines in vitro. To date, it is controversial whether ovarian carcinoma cells are capable of producing IL-10 and what effect of IL-10 in OEC.In this study, the IL-10 levels in serum and ascites of 32 patients with POEC, in culture supernatants of 4 different ovarian carcinoma cell lines and in serum of 10 patients with ovarian epithelial benign tumor and 10 health women were measured by enzyme-linked immunosorbent assay (ELLSA),and the expression of IL-10 in tissues from 64 patients with primary ovarian epithelial tumor, 35 patients with ovarian epithelial bordline tumor, 32 patients with ovarian epithelial benign tumor and 26 normal ovarian tissues was detected by immunohistochemistry(IHC). Moreover, the expression of IL-10 in three human ovarian carcinoma cell lines (SKOV3, CaoV3, and 3ao) was also detected by immunocytochemistry(ICC). Western-blot was used for detecting the expression of IL-10 in tissues and cell culture lysis above mentioned. The aim of the study to investigate the expression of IL-10 in OEC, and to interpret the relationship between peritoneal immunodeficinecyof patient with OEC and its expression of IL-10.Materials and Methods64 patients with primary ovarian epithelial tumor, 35 patients with ovarian epithelial bordline tumor, 32 patients with ovarian epithelial benign tumor and 26 normal wamen, four human ovarian carcinoma cell lines (SKOV3, CaoV3, 3ao and OVCAR) were selected for studies.
