Objective: To investigate the effects of Angelica Polysaccharide (APS) on the inducing chronic myelogenous leukemia cell into leukemic dendritic cell (CML-DC). Methods: Marrow monocytes from CML patients were cultured in GM-CSF/IL-4 or in GM-CSF/IL-4 combined with low dose (50mg/L), medium dose (100mg/L), large dose(200mg/L) or super-large dose(400mg/L) APS respectively. The morphotype of DC was identified by optical microscope or electron microscope. Dendritic cell viability were calculated by trypan blue. The phenotype of DC was identified by flow cytometry. The capability of stimulating allo-lymphocyte or auto-lymphocyte proliferation was tested by mixed leukocyte reaction (MLR). Results: Marrow monocytes from CML patients cultured in GM-CSF/IL-4 or in GM-CSF/IL-4/APS appear typical morphotype and express the high lever phenotype of DC. The capability of proliferation and the survive rate of DC were enhanced markedly and CD83, CD80 and CD86 of DC were significantly highly expressed by cultured with IL-4/GM-CSF/APS. The capability of stimulating lymphocyte proliferation was the best in the medium dose APS. Conclusion: The DC cultured with IL-4/GM-CSF/APS expressed significantly higher level of CD83, CD80 and CD86 than grown in IL-4/GM-CSF. The capability of CML-DC of stimulating lymphocyte proliferation was more potential in IL-4/GM-CSF/APS than in IL-4/GM-CSF without APS. APS can promote the inducing and mature of CML-DC.
