In spite of physiological or pathological state, the bcl-2 gene is the key to modulation. It's abnormal expression is related with many disease, expecially, with tumor. Many people think bcl-2 have the role in tumor by inhibition signal which induce apoptosis. And it may put off the aptosis which was induced by radiation,hot shock and many drug, further more, There are many proofs that bcl-2 take part in signal introduc -tion which was induced by IL-2R , and stimulate the cell prolifation by coopration with c-myc or p561ck. However, it is unknown about the bcl-2 how to act and what role. Therefore, it is key to study on the role of bcl-2.So it is important measure to construct the bcl-2 expression cell model. We construct the mbcl-2a eukanyotic expression vector, and intrduct it into cell, establish the mbcl-2ct expression cell model.Part I Construction of mouse bcl-2a gene eukanyotic expression vectorAbstract: Objective To construct the green fluorescent protein vector (pTracer-EF) carrying mouse bcl-2a gene inorder to trace the expression of mbcl-2a gene in eukanyotic cell. Methods Mouse bcl-2ot gene was inserted into pTracer EF piasmid and identified by endonuclease digestion and DNA sequencing. Then, the recombinant plasmid pTracer-EF-mbcl-2a was transfected into Cos7 cell with lipofectamine. The expression of mbcl-2a gene was verified by the use of fluorescent microscope and RT-PCR technique. Results The mouse bcl-2a gene was cloned into vector pTracer-EF and verified by PCR product and DNA sequencing. The green fluorescent protein was expressed after transfection. Conclusion the eukanyotic expression vector carrying mouse bcl-2a gene is successfully established. This research work has formed a base for monitoring the nibcl-2a gene expression and identifying protein position.
