Objective: To observe the effects of Yikunjing capsule on bone mineral density(BMD) and study its mechanism by the changes of levels of IL-6 in bone of ovariectomized rats. Methods: Adult ovariectomized rats were used in this study as the model of postmenopausal osteoporosis. 40 three-month-old of femel SD rats were randomly allocated into 5 goups: sham operation goup, ovariectomized(OVX) goup, OVX with nilestriol goup, OVX with Yikunjing groups in large and small dosage. 10 weeks after being fed with medicines, the BMD of rats' body, E2 In serum, the level of IL-6 in bone were tested, and histological changes of bone were observed.Results: (1). 11 weeks after OVX, compared with the sham OVX, the general BMD and E2 in serum decreased significantly (P<0.01) while the level of IL-6 in bone raisedobviously (P<0.01). The histological of bone revealed the trabecular became thinner and irregular. (2). 10 weeks after being fed with medicines, compared with the OVX, the general BMD in Yikunjing groups in large and small dosage and nilestriol goup increased significantly (P<0.01). No obvious change occurred in the level of E2 of nilestriol goup (P>0.05). But there were obvious raise of E2 level in Yikunjing groups in large and small dosage (P<0.05) , though the levels of E2 were still much lower than the sham OVX group. The result of bone culture revealed: the levels of IL-6 in bone decreased significantly in Yikunjing high dosage groups, the low dosage goup and nilestriol goup as compared with the OVX goup (P<0.01). The level of IL-6 in bone in Yikunjing low dosage groups was obviously higher than the sham OVX group (P<0.05) while it was lower than the OVX goup (P<0.01). Conclusions: Yikunjing capsule can effectly prevent osteoporosis in female rats induced by ovariectomy, it is able to evaluate that one of therapeutic mechanism ofYikunjing capsule was it can decrease the level of IL-6 in bone and inhibt bone absorbtion.
