| OBJECT: To investigate the ability of repairing bone defect with the compound of recombinant human IGF-I(rhIGF-I),coralline hydroxyapatite(CHA),and autogeneous red bone marrow(ARBM),and the feasibility with the compounds as bone substitute materials.METHODS:The animal models of bilateral radius bone defect(10mm in length) were created in 48 rabbits,and treated with 6 different methods respectively :A(rhIGF-I/CHA/ARBM),B ( CHA/ARBM ) ,C(rhIGF-I/CHA),D(CHA) , by E(autograft) and F(no implant) as control.At 2,4,8,12weeks,the effect were assessed by X-ray and X-ray imaginary analysis ,biomechanics,SPECT, histological studies and scoring.RESULTS:1) The bone defect were perfectly repaired with autograft and rhIGF-I/CHA/ARBM,the effect assessed by histological ,X-ray, biomechanical test,SPECT,and histological studies and scoring indicated no statistically significant difference between these two groups,and superior to the other groups with significant difference;2) The repairing effect of rhIGF-I/CHA and CHA inticated no significant difference in every observed phase.But at 12 weeks the defect were repaired with them and not with no implant.CONCLUSIONS: 1) The compound rhIGF-I/CHA/ARBM posses much high ability of rebuilding bone defect and can serve as an autograft substitute material;2) Exogeous IGF-I of low dosage alone coating to CHA has no obvious effect to promote the osteogenetic capacity of CHA in this experimental model,how to affect bone healing is expected for further studies. |
