Objective To investigate the expression of PAI-1 mRNA in early diabetic rats renal cortex and its significance. Diabetic nephropathy is one of diabetic microangiopathy in diabetes mellitus, and an important dead cause. The happen and progression of diabitic nephropathy invole several mechanism, among which the plasminogen activator inhibitor type 1 (PAI-1) appear to play an important role.Methods The renal cortex expression of the PAI-1 mRNA , urine microalbumin(MAU) and creatinine clearace ratio(Ccr) were measured in the strepotozotocin-induced diabetic rats. The STZ-induced diabetic rats were divided into four groups, eg, One week group, two weeks group . four weeks group eight weeks group. Certainly, this experience also include the normal control group. PAI-1 mRNA expression in renal cortex were determined by reverse transcription polymerase chain reaction (RT-PCR).Results The expression of PAI-1 mRNA in renal cortex is obviously increased in two groups that are four weeks group and eight weeks group (P<0.01). Compared with four weeks group, eight weeks group has distinctly diffirence(P<0.01), But MAU and Ccr only have been apparently changed in eight weeks group. The expression of PAI-1 mRNA in renal cortex are related to MAU and Ccr in four weeks group and eight weeks group.Conclusions PAI-1 mRNA expression in renal cortex of STZ-induced diabetic rats is upregulated in four weeks group and eight weeks group. This is probably related to extracellar matrix(ECM) accumulation. The expression of PAI-1 mRNA has been changed in the early stage of diabetic nephropathy. prior to the changes of MAU and Ccr.
