Objective: It is well known that hypothermia can strengthen the tolerance of cerebral to ischemic injury. To study the feasibility of resuscitation and the longest length after selective cerebral profound hypothermia and blood flow occlusion, neurological functions and essential enzymes of visceral organs were examined preoperative and postoperative, and specific protein in neurons and glial cells were examined, ultrastructure of the brains were obtained from more than one points of brain tissues.Methods: Exposed bilateral external jugular veins(EJVs), bilateral internal carotid artery(ICAs) and bilateral internal jugular vein(IJVs), a catheter was inserted into right ICA to infuse cold Ringer's solution, two catheters were distally and proximally inserted into IJV to extract the hypothermic diluted blood for ultrafiltration and then perfuse the warm blood (38℃) into the right venae cava inferior after rewarming. After both EJVs and the left ICA and the left IJV were clamped 60-80 minutes, simultaneously cold solution was immediately perfused into the right internal carotid artery to induce the brains cooling. Preoperative and postoperative changes of the serological enzyme were measured; the dysfunctional scales of neurology were done every week. At the definite date after resuscitation, the monkeys were put to death and hearts, lungs and other essential organs were examined by microscopy, and the brains were done though immunohistochemical staining for neuronspecific enlase, s-100 protein and glial fibrillary acidic protein, and the brains were examined by microscopy and transmission electron microscope.Results: In all 7 monkeys, 5 were succeeded in being set up brain-local extracorporeal circulation. After resuscitation, nervous system scores were excellent. The function of hearts, lungs and other essential organs were stable(P>0.05) after operation, NSE, s-100 and GFAP exist in the brains normally, there were no morphologic changes in all areas of the brains except for slightly swelling of the nerve fibers in the oblongata.Conclusions: It is safe that monkey resuscitated from selective cerebral deep hypothermia after blood flow occlusion 60-80min, and there is no neurological deficits and no damage to each essential organs, the ultrastructure of the brains is normal and the conformation of every organelles is natural, immunohistochemical staining of brains is normal. That is to say that DHBO has great protective effect on cerebral ischemia.
