| Objective: To explore the role of ANG in angiogenesis of gastric cancer.Methods: ANG mRNA expressions of 68 gastric cancer tissues and paired adjacent gastric mucosas were detected by reverse transcript polymerase chains reaction (RT-PCR). The expressions of CD34 , Ki-67, VEGF and ANG of the same gastric cancer tissues were observed by immunohistochemistry. Microvessel density (MVD) were figured out according to their expressions of CD34. The proliferation indexes of cancer cells of gastric cancer tissues were figured out according to their expressions of Ki-67. Results:1.The expression of ANG mRNA was found both in gastric cancertissues and their paired adjacent gastric mucosas. In the 68 cases, theANG expression level of gastric cancer tissues (0.482±0.094) issignificantly higher than that of the paired adjacent gastric mucosas(0.276 ± 0.019 P=0.03) .2. Ratio of the ANG mRNA expression of the gastric cancer tissue and the paired adjacent gastric mucosa (T/N) is significantly related to the MVD. With the increase of ANG mRNA expression, MVD elevates. A significantly positive correlation between them is found (r=0.380, P=0.001) The ANG expression in gastric cancer tissues increases with the expression of ANG mRNA, and a positive correlation is found between them. With the increase of ANG expression of the gastric cancer tissues, MVD elevates,and a positive correlation is found between them.3. The ANG expression in the gastric cancer tissues is significantly related to the expression of vascular endothelial growth factor (VEGF). With the increase of ANG expression of the gastric cancer tissues , VEGF expressoin elevates. There is a positive correlation between them(P=0.009) .4. The mean MVD of the group whose VEGF and ANG are both positive is significantly higher than that of the group whose VEGF and ANG are both negative, the difference is significant (P=0.0267) .5. The ANG expression in the gastric cancer tissues is significantly related to the proliferation index of cancer cells in gastric cancer tissue. With the increase of ANG expression in gastric cancer tissue, the proliferation index of cancer cells of gastric cancer tissue elevates. There is a positive correlation between them (P=0.009) .6. The expressions of ANG mRNA, ANG and VEGF in gastric cancer have no relation to age, location of the tumor, histological type, lymph node metastasis and clinical staging of the patients.Conclusions:1. The ANG expression in gastric cancer tissues is higher than that of the paired adjacent gastric mucosa.2. In gastric cancer, there is a positive correlation between ANG expression level and MVD. ANG acts as an angiogenic factor which promotes the growth of the carcinomatous blood vessel.3. ANG may work in coordination with VEGF in promoting the angiogenesis of gastric cancer.4. There is a positive correlation between ANG expression and tumor cell proliferation index in gastric cancers. ANG may have the function to promote the growth of the cancer cells.In a word, this study helps to further understanding the mechanism on angiogenesis of gastric cancer and provides theoretical reference to explore a new way of antiangiogenesis in gastric cancer. |
