Objective : To evaluate the effects and its possible mechanism of propofol on acute renal ischemia-reperfusion injury in spontaneously hypertensive rats(SHR).Methods : Model of acute renal ischemia-reperfusion injury was made by placing the atraumatic vascular clamp in renal pedical for 60minutes.Seventy-five male SHR at age 12-14 weeks were randomly allocated into three groups ,each group has twenty-five rats (n=25) ,false opertaing group(A) ,ischemia-reperfusion group (B) and propofol-group(C).Each group has five observative time points: 10 min before ischemia(Io),ischemia for 60min (I ),lhour after reperfusin (R1),3hour after reperfusion (R2),6hour after reperfusion (R3).Each point has five rats .In groupC,propofol 20mg/kg was given intravenously 5min prior to clamping and infused at a rate of 60mg/kg/h for 60min .Other groups of rats received normal saline as controlled ,but the renal pedical was not occlused in groupA.The parameters including BUN,Cr,MDA,Ang-II,IL-8 and B2-MG were observed before,during ischemia and after reperfusion.The kidney specimens were eximined using light microscopy, the Pallers score for acute renal tubular injury.Results : 1 During the ischemia-reperfusion In groupB serurm BUN.Cr.MDA .urine 02-MG levels and the Paller' s score for acute renal tubular injury increase markly compared with those in groupA and before ischemia(p<0.05); In group C serurm BUN,Cr,MDA, urine pVMG levels and the Paller' s score for acute renal tubular injury decreased markly compared with those in group B(p<0.05).2 IL-8,Ang-II,ET concentration after reperfusion were significantly decreased in group C compared with those in group B (p<0.05).3 NO concentration decreased significantly in group B compared with those in group C(p<0.05).Conclusion: 1 Ischemia-reperfusion injury in SHR kidneys possibly was effected renal cortex and serum active substance such as NO,ET, Ang-II,IL-8.2 Propofol can effectively protect SHR kidneys ischemia-reperfusion injury. 3 It shows that propofol may prove to be useful in prevention of ischemia-reperfusion injury in hypertensive pataints and improvement of renal operation prognosis.
