| Object To investigate the effect of taxol on vitro invasion, expression and activity of gelatinase (MMP~2, MMP-9) of human adenoid cystic carcinoma(ACC). Methods Highly mestastatic ACC cell line(ACC-M) was adopted, which was treated with various concentration of taxol (0.1, 0.06, 0.01mg/L). Using MTT technique, to assay the ACC-M cell-ECM adhesion and the effect of taxol on viability of ACC-M. ACC-M cell-ECM invasion and migration were evaluated using modified Boyden chamber. Using technique of colorless lot, to assay the activity of gelatinase (MMP-2, MMP-9) in culture supernatant of ACC-M. The contents of MMP-2 and MMP-9 in culture supernatant was detected with ELISA. In addition, to evaluated the suppression of taxol on expression of MMP-2 and MMP-9 of ACC-M by immunocytochemistry (LSAB method). Results It was found that taxol (0. 1-0.01mg/L) inhibited ACC-M to adhere and migrate by 13. 3%-41. 2%, 22.0%-72.3%, respectively. At concentration of 0.1mg/L, 0.06mg/L and 0.01mg/L, taxol suppressed ACC-M to invade the ECM by 78.4%, 64.9%, 42.3%, respectively. In addition, the effect of taxol on viability of ACC-M was negligible. The diameter of each colorless lot indicated the activity of gelatinase.It was found that taxol(0. 1-0. Olmg/L) inhibited activity of gelatinase by 66. 6%-38. 8%. At taxol concentration of Omg/L, 0.01mg/L, 0. 06mg/L and 0. Img/L, the contents of MMP-2 in culture supernatant were 581pg/105 cell, 397 pg/105 cell, 187 pg/105 cell, 86 pg/105 cell, the contents of MMP-9 were 633pg/105 cell, 273 pg/105 cell, 208 pg/105 cell, 127 pg/105cell. In addition, it was showed that positive indexes of expression of MMP-2 and MMP-9 of ACC-M were decreased remarkably following the increasing of taxol concentration. Conclution Taxol can significantly inhibit vitro invasion of ACC-M. In addition, taxol can result in the down-regulation of expression and activity of MMP-2 and MMP-9. Therefore, It can be deduced that taxol can exert multiple suppressive effects on the invasion and mestastasis of adenoid cystic carcinoma. |
