| Objective: This study is to explore the effect of TGF- β2 on rabbit retinal pigmentepithelial(RPE)cell proliferation and DNA synthesis.On the basis of previousresults,we can reveal the effect of TGF- β2 on the formation and development ofproliferative vitreoretinopathy(PVR).Methods:First of all,establish rabbit RPE cell line to make primary cell culture.Dividecells into two groups:blood serum group and non-blood serum group.Adds. 10μMlysophosphsphatidic acid(LPA), 2ng/ml TGF-β2 and LPA+TGF-β2 to intervenecell growth progress respectively.Investigate the results of cell proliferation and DNAsynthesis.Results: 1.Either 10μM LPA alone or in coordination with blood serum can exertauxo-action influence on RPE cell proliferation.2.TGF-β2 can inhibit RPE cell proliferation and DNA synthesis.Inhibitive effect ofTGF-β2 alone is more significant than LPA+ TGF-β2.3 .TGF-β2 can inhibit RPE cell proliferation and DNA synthesis owing to LPA'sreinforcement.Conclusions:RPE cell plays an important role in the occurrence and progress ofvitreoretinopathy .Application of TGF- P 2 can provide a new way to prevent,controland treat PVR,while application of LPA can provide a new way to treat degenerationdisease with respect to RPE cells. |
