Objectives:To investigate the expression of c-fos oncogene in rats central nevous system following Isoflurane anesthesia,research Isoflurane action sites in CNS,and discuss general anesthetic mechanisms of inhalation anesthetics. Methods:Forty adult SD rats,weight 178.6±18.0g,were randomly divided into four groups:control group(n=10),0.6% Isoflurane group(n=10),1.2% Isoflurane group(n=10),1.8% Isoflurane group(n=10).Experimental rates were inhalated 0.6%, 1.2%. 1.8% Isoflurane for 1 h.The control groups rats underwent the same steps except without inhalating Isoflurane.After 1h.each rat was anesthetized by intraperitoneal injection Pentobarbital(40mg/kg),opened rats chest and perfused normal saline 100ml as well 4% poly-formalindehyde 250ml.90 min later the whole brain and L4-6 section of spinal cord were removed and paraffined-inbeded and insected.C-fos oncogene immunohistochemical technique was employed.To observe and count on the Fos-like immunoreactive(FLI) cells. Results:Control group didn't find FLI cell.The experimental groups could observe FLI cells in the cortex, thalamus, midbrain, bridgebrain. medulla oblongata and cerebellum,compaired with control group p<0.05 and p<0.01.those FLI cells were limit in someregion. Meanwhile finding FLI cells were increase with Isoflurane concentration raised,within experiment group compared p<0.05 and p<0.01.In spinal cord,0.6% Isoflurane group didn't find FLI cell. 1.2% group observe FLI cells in posterior hom,compared with control group p<0.01,1.8% group appeared more and more FLI cells in posterior horn,even anterior horn,compared with control group and 1.2% group p<0.01. Conclusions:;(1) Isoflurane can induce expression of c-fos oncogene in the cortex,thalamus, midbrain, bridgebrain,medulla oblongata,cerebellum and spinal cord of rats.(2) The cortex,thalamus, midbrain, bridgebrain,medulla oblongata,cerebellum and spinal cord are the action sites of Isoflurane.(3) 0.6% Isofluranes anesthetic sites is mainly super-spinal-cord central nervous system...
