OBJECTIVE To master the culture method for inducing human acute myeloid leukemic cells to differentiate into dendritic cells. Dendritic cells pulsed with antigen peptides in two different ways, observing the effect of different antigen peptides pulsing manners on the activation of autologus T lymphocytes. To provide a new vitro study method for immunotherpy of leukemia.METHODS Get dendritic cells by cultured plastic adherent monocytes isolated from acute myeloid leukemia patients of peripheral blood with granulocyte-monocyte clony stimulating factor(GM-CSF) and interleukin-4 (IL-4) and tumor necrosis factor-a (TNF- a ) for 7-14 days. Dendritic cells were recognised using methods including mophology , immunphenotype and the ability of processing and presenting antigen to T lymphocytes.Leukemic antigen peptides were obtained by freezing and thawing repeatedly. Dendritic cells were pulsed with antigen in manner of leukemic antigen peptides and antigen peptides-Hsp 90 compounds respectively. Useing the dendritic cells presenting antigen peptides to autologus T lymphocytes, the cytotoxity of actived T lymphocytes to autologus leukemic cell were measured in vitro using the lactate dehydrogenase-release assay.RESULTS We got dendritc cells from monocytes isolated from three patients' peripheral blood. After pulsing with leukemic antigen peptide or leukemic antigenpeptide-Hsp90 compounds, autologus lymphocytes cocultured with AML-derived dendritic cells were able to lyse autologus leukemia cells, the cytotoxicity values of the T lymphocytes were 60.3 7.7%and 32.9 8% respectively. CONCLUSIONS Human acute myeloid leukemic cells can be induced into dendritic cells in the culture system, but the ability to differentiation among patients is heterogeneity. When pulsing dendritic cell with leukemic antigen peptide-Hsp90 compounds, the autologus T lymphocytes had the stronger effect on cytotoxicity of autologus leukemia cells than the simple antigen peptide's.
