OBJECTIVE : To investigate the antinociceptive effect of systemic administration of NMDA receptor antagonist , Ketamine and sciatic nerve block for model of peripheral neuropathic pain in rat .METHODS : At anesthetic condition , 32 healthy male rats were made into chornic contriction injury to the right sciatic nerve ( CCI model ) ; then were randomly divided into 4 groups , 8 rats of each . The group A was control group , group B,C,D was experimental group . After fourth postoperative days , the rats of group B were injected Ketamine 0.3mg/kg , 0.5ml by retail vein ; the rats of group C were made right sciatic nerve block with 1.0% Lidocaine 1ml ; the rats of group D were injected Ketamine 0.3mg/kg , 0.5ml by vein and combined the right sciatic nerve block with 1.0% Lidocaine 1ml ;the treatment was practiced once everday throughout seven days. And everyday the rats of each group weretested spontaneous neciceptive behaviors and thermal struggle latency of hindpaw immersion into a water bath ( 46 ℃ ) until tenth days .RESULTS : There were no difference at rats' weight, volume of anesthetic, the spontaneous nociceptive behavior and thermal stimuli and after surgery between each group (P>0.05)。Compared with group A , the values of spontaneous pain score and thermal stimuli test of B,C,D group were different frome fourth to tenth postoperative days (P<0.05);the values of spontaneous pain score and thermal stimuli test were no diffience between group B and C (P>0.05);the values of spontaneous pain score and thermal stimuli test of group D were different from group B,C (P<0.05)。CONCLUSIONS : The NMDA receptor antagonist , Ketamine and sciatic nerve block were all effective for neuropathic pain in rats , but the NMDA receptor antagonist , Ketamine combined with sciatic nerve block was a more effective and promising approach in the treatment of neuropathic pain .
