| Objective:In the study,we established the peripheral blood stem cell transplantational(PBSCT) murine model. Methods:Balb/c mice, 120 female mice were served as recipients, 60 male mice were served as donors. Each male mouse was intra-abdominally injected with CTX400mg/kg 6 days before transplantation, from the fifth day each male mouse was subcutaneously injected with G-CSF5 u g once, twice a day till the day before transplantation. A Cs137 source was used for total body irradiation (TBI) of the recipients on the transplantation day. The nucleated cells were collected under sterile conditions by dissection of male mice eyeball blood. 120 irradiated mice were randomized into four groups. In four hours after irradiation , three groups of them were injected with nucleated cells respectively cells per mouse via caudal vein,the other group as control group was injected with the same volumn normal saline(NS) .Then we recorded the changes of numbers of peripheral blood leucocyte in transplanted mice , the effects of different cell numbers to survival rate of recipients ; the condition of spleen colony formation and the proportion of Y chromosom in recipients. Results: Onthe 7th day after peripheral blood stem cell transplantation the numbers of leucocyte were the lowest.This condition was different from bone marrow transplantation model.From the 7th day the numbers of leucocyte recovered gradually but on the 15th day they were still quite lower than normal number.After transplantation the spleen colony on the 9th day was quite few than on the 12th day, p<0.01.On the 12th day the spleen colony of the 5 X 106 group and 1X 107 group was quite more than that of the 1 X106 group, p<0.01.We assayed the number of specific Y chromosome in recipients by means of C-zone at the third month after transplantation. The results showed that the percentage of cells whose chromosome were XY among 50 cells was 26.25+4. 42% . Conclusion: The model of peripheral blood stem cell transplantational murine has been successfully finished. The establishment of this model will be beneficial to the relative basic research of PBSCT. |
