A liposome preparation method called ethanol injection was adopted to prepare Propranolol hydrochloride-containing liposome in this paper. In order to ensure excellent stability and to obtain higher entrapment efficiency, a uniform design was carried out and obtained the optimum component of liposome and the entrapment efficiency was above 40%. In accelerated tests, it was found that the liposome suspension was stable at low temperature in oxygen-free atmosphere protecting from light.A method was developed to determine the entrapment efficiency of Propranolol hydrochloride-containing liposome preparations by hydrochloride acidification , chloroform extraction-UV spectrophoto-metry and gel filtration chromatography-UV spectrophotometry. The method was simple and accurate and easy to operate.The physicochemical properties such as size distribution, entrapment efficiency, viscosity, surface tension of liposomes and nebulized liposomes were studied and the results indicated that the nebuli-zation had an important effect on these . For example, the mean size was decreased from 253. Inm to 209. 4nm during the nebulization; the viscosity and the surface tension became higher after nebulization. The ciliary toxicity of Propranolol hydrochloride liposome nasalABSTRACTpreparation was investigated by using index of ciliary vibration duration by in situ method. The conclusion was that the liposome preparation had less toxicity than its solution , for example it prolonged the ciliary vibration duration from 5 minutes to 250 minutes. It had been found that the preparation had no irritation. LD50 i.p. in mice of the liposome preparation was 153.1mg/kg. All above results indicated that the liposomal preparation markedly decreased the toxicity of the drug.The effects of Propranolol hydrochloride-containing liposome preparation on epinephrine-induced arrhythmia in conscious rabbits revealed that the liposome preparation markedly reduced the duration of arrhythmia from 191 seconds to 79 seconds and was as effective as i.v. administration.
