Heat shock proteins(HSPs), also named stress proteins, are a group of cellular housekeeper proteins. They are constitutively expressed in all cells in the physiological conditions to maintain normal cellular form and function. The aim of this experiment is to establish the correlation between the infection of human papillomavirus and the expression of HSP70 in vulvar leukoplakia.Methods :In order to find the role of HSPs in the pathogenesis of Genitalia Leukoplakia, we detected the expression of HSP70 in epidermal keratinocytes with sections from formalin-fixed and paraffin-embedded blocks of biopsy specim ens from Genitalia Leukoplakia lesion (80 cases), squamous cell carcin oma (48cases) and normal skins(20 cases) by means of immunohistochemical LSAB method. And we measured the expression of HSP70 in epidermis so as to study dynamic state of occurrence and recovery on Genitalia Leukoplakia lesion. The infection of HPV in specimen in which (he HSP70 was positive was studied.Results: Our results showed that: the expression of HSP70 among the normal skin group and Genitalia Leukoplakia groups were significantly different(P (0.05 ; and the difference of expression of HSP70 was not significant among three groups (P>0.05). Compared with the squmous cell carcinoma group, the differences were highly significant(P<0.05). the difference are not significant in different grade of squmous cell carcinoma(P>0.05). In epidermal keratinocytes of normal skins-, Genitalia Leukoplakia and squmous cell carcinoma, the expression of HSP70 is regularly increased from basal layer to granular layers with cytoplasmic staining pattern, the differences were highly significant(P<0.05). HPV-DNA was increased in the Genitalia Leukoplakia in which HSP70 was positive.Conclusion: Our study indicated that epidermal keratinocytes in which the expession of HSP70 was increased not only do their effect on the normal proliferation and differentiation but also their stress protective function in the Genitalia Leukoplakia.
