Contusive eye injuries can cause many eye complications, such as anterior chamber angle recession , hypertension , corneal abnormalities and cataract and so on. Injured cataract is one of the most important complications, with the morbidity of 36%-52%, induced by the perforating or contusive wounds in eyes, which is also the leading cause of the blinding, about 28.2% of all the visual disturbances. In 2001, Viestenz analysed 417 cases of contusion and rupture of the globe with frequent avoidable causes of trauma in theErlangen Ocular Contusion-Registry from 1985 to 1995 and reported that ocular trauma was the cause of every third enucleation, which blunt injuries belonged to the most frequent causes. Also in 2001, Machimoto Tadashi established the contusion cataract model of C57/BL10 mouse. But till up now it was rarely known for the mechanism of contusion cataract, so was the accurate report for the ultrastructure of lens epithelial cells.The purpose of this study is to find one way, which is more quickly and more credible, to establish the typical animal model (rats) of contusion cataract for discussing the mechanism of contusion cataract and to detect the ultrastructure of lens epithelial cells to discuss the relationship of cell apoptosis and contusion cataract formation.1. Animal model of contusion cataract50 SD rats (female, four weeks old) were divided into five groups randomly and all general anesthetized. Group A: to hit left eyes of the rats with a 10g ball from 10cm height continuously for 100 times; Group B: to hit left eyes of the rats with a 10g ball from 20cm height continuously for 100 times; Group C: to hit left eyes of the rats with a 20g ball from 20cm height continuously for 100 times; Group D: to hit left eyes of the rats with a 20g ball from 20cm height continuously for 100times per week, five weeks totally; Group E: to hit left eyes of the rats with a 30g ball from 20cm height continuously for 100 times per week, five weeks totally. Then to observe the changes of the lens of rats with a slit-lamp microscope. After one week, different changes appeared in the experimental lens of the five groups. But after thirty days, the changes disappeared in Group A. The opacifications still appeared in group B , C and D, only in D group were there vossious opacifications. In Group E there were other diseases caused by contusion. The quantitative analysis results show there were no significant differences between the B and C groups (10g:20g)(p=0.858>0.01), and significant differences between the C and D groups (100:100x5)(p=0.018<0.05). All the above suggested that typical and stable cataract could be induced quickly by the way of D group (to hit left eyes of the rats with a 20g ball from 20cm height continuously for 100 times per week, five weeks totally ) . So the way of the D group was recommended to be used for establishing the rats model of the contusion cataract.2. The ultrastructure of lens epithelial cells of contusion cataractAfter the opacifications appeared, the rats were generalanesthetized, anterior capsules were got to make the specimenfor transmission electron microscope (TEM) to observe the ultrastructural changes of the lens epithelial cells. The other eye was served as self-control. Among Group B, C and D , the characteristic apoptosis cell morphological changes can be seen only in the experimental lens epithelial cells of the Group D. No obvious changes can be found in the self-control eyes. All the above suggested that lens epithelial cells apoptosis is related with contusion cataract formation.The results above suggested the contusion injures are the unquestionable cause of the complicated cataracts. All the above also suggested that typical and stable cataract could be induced quickly by the way of D group (to hit left eyes of the rats with a 20g ball from 20cm height continuously for 1 00 times per week, several weeks totally) ,so this way was recommended to be used for establishing rats model of the contusion cataract.
