Objective: To detect the expression of SAMDC-mRNA in the cortex and subcortex of rat after the different time of the reperfusion following 2h focal cerebral ischemia by occlusion of MCA in order to explore the variety and signification of SAMDC-mRNA in this condition.Methods: 2 hours of reversible focal ischemia in rats were produced by improved the method of Connolly' suture occlusion of MCA and the expression of SAMDC-mRNA in the cortex and subcortex was measured by RT-PCR after 2,4,8 and 24h reperfusion following 2h ischemia respectively. The time courses of SAMDC-mRNA expression in the experimental and control group were analyzed.Results: l.There was no expression of SAMDC-mRNA in the control group. But in the experimental group, the expression in the side of the MCA occlusion has arisen two times. One showed after 4h reperfusion, and it was significant compared with 2h and 8h reperfusion (P<0.01). Another appeared after 24h reperfusion. It was the peak value and there also was difference compared with 4h reperfusion significantly (P<0.01). 2. The expression of SAMDC-mRNA in the contralateral cortex and subcortex was increased obviously after 8h reperfusion and was decreased after 4h, 24h reperfusion. There was no expression of SAMDC-mRNA expression during the ischemia and after 2h reperfusion.Conclusion: 1. Ischemic Model of MCA in Rat can be copied effectively and easily by improved the method of Connolly's suture occlusion. 2. The change of SAMDC-mRNA expression was biphasic that has arisen two times in 4h, 24h after reperfusion respectively.
