[Abstract] Objective: On the base of steroid-induced Adipogenesis in Marrow stromal cells, to oberve the influence of pravastatin and comment on the machicm of influence. Methods: The osteoprogenitor cells of the femur of the rabbit were procured and cultured, after adherent growth culture in vitro ,The samples were isolated and were treated with dexamethasone which acted as adipocyte ditterentiation inducer and pravastatin..On the one hand .The cells in culture were stained with Sudan III,The number of adipocytes were counted on under a light microscope. On the other hand, The Alkaline phosphatase of the cells were measured. Results: On the base of 1.0 10-6 concentrations of dexamethasone, the cells in culture were treated with increasing concentrations of pravastatin for 21 days ,The number of adipocyses which in 6.4 10-3mmol/ml concentration of pravastatin were lower than the control, decreased with higher concentration of pravastain and showed a dose-dependent relation..Alkalin phosphatase activity in the cultured cells which were treated with pravastatin were higher than that in the control..Conclusion: Dexamethason can directly induce marrow stromal cell differentiation into a larger number of adipocytes and inhibit osteogenic differentration; but pravastatin can inhabit the function of dexamethsaon. Which procide a way to treat andprotect necrosis of femoral head that was induced by dexamethason.
