Objective To study the effect of Danshen on action potential (AP), L-type calcium current (Ica-t)' ATP sensitive potassium current (IK(ATP)) and transient outward potassium current (Ito) in isolated cardiomyocytes after ischemia, investigate the mechanism of Danshen in prevention and treatment of myocardial ischemia. Methods Using the whole cell patch-clamp technique in ischemic single cardiomyocyte enzymatically isolated from rabbit ventricle. Results (1) Danshen remarkably shortened the action potential duration (APD ) both normal and ischemic cardiomyocytes; Danshen had no effect on resting potential (RP), action potential amplitude (APA) and Vmax . (2) Danshen remarkably reduced Ica-t in dose-dependency. (3) Danshen increased Ito and IK(ATP> of ischemic cardiomyocytes, respectively. Conclusion Danshen decreased Ica-L and shortened APD of ischemic cardiomyocytes, which may play a role in the mechanism of anti-myocardial ischemia and ischemia induced arrhythmia.
