The nucleoprotein of rabies virus is responsible for encapsieation of viral RNAs, and the L protein , in cooperation with the P protein function as an RNA-dependent RNA polymerase. And N protein alone can induce protective immunity against lethal infection in animals. Furthermore, the homology of nucleoprotein genes derived from different rabies virus strains is rather conservative. As a result , it is widely used the molecular epedemiology in the world -. rabies virusdetection and genetic vaccine.The purpose of cloning and sequence analysis of nucleoprotein genes of rabies virus 8202strain is identifying its genotype and researching efficient genetic vaccine . The nucleoprotein genes(about 1400nt) of strain 8202,isolated from deer, ranging from 69nt to 1463nt were amplified by RT, semi-nested PCR. They were cloned into PUCm-T vector and sequenced by biology company. Comparison and analysis of the nucleotide sequence and amino acid sequence deduced with that 7 strains previously published were performed by computer with DNAsis software.Results showed that Ag I ,III, IV sites exited different strains was exactly common except Ag II site.Q79 was changed into E79 at Ag II site of 8202.lt is possible that the only acid changed will affect the secondary structure N protein of 8202 strain. Phylogenitic trees were established. The nucleotide and amino acid homology of 8202 with 3aG were 93.9% and 94.6% respectively. But the homology with other strains was also more than 90.0%. It proved that all of strains including 8202 strain were genotype I and strains 8202 and 3aG were divided into sub-genotypes.
